Studies on the development of a vaccine against Mycobacterium sp

چکیده
Mycobacteriosis, caused by Mycobacterium sp., results in severe loss of fish production in Japan’s aquaculture industry. Since the bacterium can survive and grow in side host cells, chemotherapeutic agents are not effective and potent vaccines are required to control the disease. In this study, the effects of two vaccine candidates, Bacillus Calmette and Guèrin (BCG, an attenuated strain of Mycobacterium bovis) and formalin-killed cells (FKC) of Mycobacterium sp. strain 012971, on the immediate and acquired immune response were evaluated in Japanese flounder, Paralichthys olivaceus. On 1, 3 and 7 days post treatments with these candidates, gene expression levels of inflammatory cytokines encoding interleukin-1β (IL-1β), IL-6 and tumor necrosis factor α (TNFα) increased. This suggests that the vaccine candidates stimulate immediate immune responses. To determine if specific defenses against mycobacteriosis were acquired, purified protein derivative (PPD) from Mycobacterium sp. strain 012971 was injected intramuscularly into fish at 4 weeks after vaccine treatments and the inflammatory cytokine gene expression levels were analyzed. Gene expression levels of the inflammatory cytokines were increased only for the fish treated with BCG, but not those with FKC. Subsequently, the vaccinated fishes were challenged with Mycobacterium sp. The relative percentage survival (RPS) of BCG and FKC vaccinated groups at 12 days post challenge were 37.5 and 18.1, respectively. These data indicate that BCG might be useful as a vaccine against fish mycobacteriosis.

Background: Tuberculosis (TB) remains as a major cause of death around the world. Construction of a new vaccine against tuberculosis is an effective way to control it. Several vaccines against this disease have been developed. The aim of the present study was to cloning of tb10.4 gene in pcDNA3.1+ plasmid and evaluation of its expression in eukaryotic cells. ...
متن کاملExtraction and biological evaluation of Mycobacterium kansasii extracellular vesicles as a vaccine candidate against mycobacterial pulmonary infections
Introduction: Extracellular vesicles (EVs) are bacterial products with diverse biological roles. Like many microorganisms, Mycobacterium kansasii as a nontuberculous mycobacteria (NTM), can naturally release EVs. The aim of the present study was the extraction and biological evaluation of M. kansasii as a vaccine candidate against mycobacterial pulmonary infections. Methods: After bacterial cul...

The identification of a large number of antigens with potential for development of new tuberculosis vaccine has been accomplished in recent years. This study was designed for cloning and expression of ESAT-6 as a potent antigen of Mycobacterium tuberculosis. Selected gene (Rv3875) was amplified by PCR and product was ligated into expressing plasmid vector pQE30 and recombinant pQE30-ES plasmi...
متن کاملA study on the immune response induced by a DNA vaccine encoding Mtb32C-HBHA antigen of Mycobacterium tuberculosis
Objective(s): Tuberculosis (TB) has still remained a global health issue. One third of the world's population is infected with tuberculosis and the current BCG vaccine has low efficiency; hence, it is necessary to develop a new vaccine against TB. The aim of the current study was to evaluate the efficiency of a novel DNA vaccine encoding Mtb32C-HBHA antigen in inducing specific immune responses...

Background & objective: Tuberculosis (TB) remains a major cause of death around the world. Bacillus Calmette Guérin (BCG) is the only vaccine used in TB prevention that has a protective effect in children, but its effectiveness declines in adults. Design and development of new vaccines is the most effective way against TB. The aim of this study was to design and construc...
